One-step Lactoferrin Purification

Lactoferrin purification from crude sweet whey in one step, an expanded bed adsorption process was proposed with cation-exchanger Bettsep® SP700.

Lactoferrin (Lf) is an 80 kDa iron-binding glycoprotein with multifunctional properties and useful for clinical and commercial applications. Currently,  Lf has some advantages including, immune system modulation antibacterial activity and as an antioxidant in infant and adult of human as well as animal health. In the present study. Lactoferrin is considered to be an important host defense molecule and has a diverse range of physiological functions such as antimicrobial/antiviral activities, immunomodulatory activity, and antioxidant activity.

The adsorption equilibrium and kinetics of lactoferrin with Bettsep® SP700 were measured. The results indicated that Bettsep® SP700 had fast adsorption of lactoferrin and was suitable for high-velocity operation. The elution condition was optimized as 1 M NaCl in the phosphate buffer (pH 7.0).

Finally, lactoferrin was successfully recovered with a high purity of 99.7% in a single step.
